DOĞU MAZISININ (Thuja orientalis L.) EMBRİYO KÜLTÜRÜ İLE ÇOĞALTIMI

Cupresseaceae familyasına ait Doğu mazısı (Thuja orientalis L.), peyzaj açısından kullanımıyla birlikte kereste ve odun üretimi için önemli bir orman ağacı türüdür. Bununla tıbbi olarak kullanımı mazıyı oldukça bitkiler sınıfına sokmaktadır. Çalışmada, bitki biyoteknolojisinin tekniklerinden biri olan embriyo kültürü tekniği ile çoğaltarak sonuçlar elde edilmiştir. Sakarozun farklı dozları birlikte, Woody Plant Medium (WPM) temel besin ortamına 6-benziladenin (BA)’in (0, 1, 2 mg l-1) 2,4-Diklorofenoksi asetik asit (2,4-D) veya Indol-3-bütirik (IBA)’in 0.5 1 kombine edilerek ilave BA (1 + IBA (0.5 büyüme düzenleyici madde (BDM) kombinasyonuna 30 g/L sakaroz edilmiş ortamda çimlenme başarısı %100 oranında meydana gelirken, maksimum sürgün uzunluğu (4.8 cm) kök sayısı (7.6) 2,4-D BDM 45 eklenmiş ise (2.4 belirlenmiştir. Aklimitizasyon uygulaması yapılan bitkilerden %69’u dış koşulara uyum göstermiştir.
